Lab 3: ESTIMATION OF CREATININE
Objective:
To perform the Jaffe method in creatinine estimation proper procedure, and the way to get reliable result.
Introduction:
Creatinine is a break down product of creatine phosphate in muscle, and is usually produced at a fairly constant rate by the body (depending on muscle mass). Creatinine is chiefly filtered out of the blood by the kidneys, though a small amount is actively secreted by the kidneys into the urine.the creatine is used to estimate the healthy of the kidney because when the creatine level down it means the kidney are having problem
Principle:
Protein in serum will be deposited by tungstic acid and then the deposit is separated by centrifugation. The supernatant creatine will be reacted with red picric acid. This reaction is called Jaffe reaction. The color formation produced is measured by 520 spectrophotometer.

Discussion:
The Creatinine is a chemical waste product that's produced by your muscle metabolism and to a smaller extent by eating meat. Healthy kidneys filter creatinine and other waste products from your blood. The filtered waste products leave your body in your urine.
If your kidneys aren't functioning properly, an increased level of creatinine may accumulate in your blood. A serum creatinine test measures the level of creatinine and gives you an estimate of how well your kidneys filter waste (glomerular filtration rate). A creatinine urine test can measure creatinine in your urine.the sample that can be used either serum or the urine.but in this experiment we used the serum for the creatine test.3 sample test tubes are tested their absorbances as the blank,test and standard.
We used the working standard solution creatine 10 mg/dl,this means the equation to calculate the result is times by 10.the working solutions can be different by any value but it not affect the normal range so that don’t be scared to use any type of working solution on different value.the result that have been get will be compare on the references value to indicated the patient are normal range or not.by using it we can know the condition of the patient.
Normal values can vary from laboratory to laboratory, but are generally in the following ranges:
· Adult female: 0.5-1.1 mg/dL
· Adult male: 0.6-1.2 mg/dL
· Adolescent: 0.5-1.0 mg/dL
· Child: 0.3-0.7 mg/dL
· Infant: 0.2-0.4 mg/dL
· Newborn: 0.3-1.2 mg/dL.
A creatinine clearance test includes obtaining a blood sample (for serum creatininedetermination) AND timed urine specimens (for urine creatinine determination). Both are needed to compute (just google the formula) for the the creatine clearance, defined as a calculated measue of glomerular filtration rate, reflecting how well the kidneys remove creatinine from the blood.Creatinine clearance test therefore has the advantage of determining, of course, the creatinie clearance, versus a serum creatinine alone though its also helpful knowing that renal disease is the only pathological condition that raises the serum creatinine level.
The disadvanyege are the only active person can be seen it color changes of the solution that have been made for tested.the oreange color,it can be affected by the drug and medication of the person.
Conclusion:
In this experiment, the students will be too able to perform the Jaffe method in creatinine estimation following a proper procedure, and manage to produce reliable result.
Questions:
1. Apart from measuring creatinine in urine, name other particles that can be detected in urine to determine normal kidney function?
Urea because urea is a by- product from metabolism of proteins by the liver and is removed from the blood by the kidneys. If any increase and decrease urea in urine indicate has problems to renal itself.
2. In your opinion, why creatinine is the most ideal indicator to determine normal kidney function?
In my opinion, creatinine is a chemical waste molecule that is generated from muscle metabolism. If abnormally high levels of creatinine thus warn of possible malfunction or failure of the kidneys. It is for this reason that standard blood tests routinely check the amount of creatinine in the blood.
